As with any biological system protein pathways are complex with numerous chemicals that act to increase or decrease the amount of protein produced and how it behaves in the body. The result is an interconnected web of protein pathways that are not fully understood. One of the most common sites of action of Covid 19 is the lungs thought to be due to the presence of ACE2 protein in cell membranes of lung cells. Let’s focus on the lungs and see what happens as it’s the most common cause of severe symptoms.

After entering the cell and replicating totally un-checked infected cells burst open releasing more virus to infect other surrounding cells. A process of positive feedback increases cell damage and functioning of the gas exchange mechanism in the lungs.
The protein pathway mentioned above, which the virus targets, is the renin-angiotensin system involved in regulating blood pressure in various parts of the body. At the lungs activation of certain parts of this pathway have bad affects including vasoconstriction of the small blood vessels involved in the mechanism of gas exchange in the lungs. It also increases inflammation of alveolar cells and fibrosis which can produce chronic reduction in lung function.
Acute inflammation of lung cells are seen in pneumonia and acute respiratory distress syndrome (ARDS) both exacerbated by Covid 19. As a comparison chronic inflammation is represented by asthma and chronic obstructive pulmonary disease (COPD).
Due to the affects of inflammation, cell debris and junk proteins thrown out into the surrounding area due to cell death the blood flow to the lungs gets cluttered. Tiny clots form in the blood vessels involved in the gas exchange mechanism of the lung. Reducing the uptake of oxygen from the air deep in the lungs.
Pulmonary hypertension results from constriction of the blood vessels involved in the gas exchange mechanism of the lungs. Resulting in impaired oxygen uptake within the lungs. Pulmonary hypertension results from higher levels of Angiotensin 2: a protein produced in the renin-angiotensin system. Increased Angiotension 2 results in reduced blood flow to the lungs.
To summarise briefly, there are three causes of reduced lung function. Death of Alveoli cells deep in the lungs, tiny blood clots in the vessels supplying the gas exchange mechanism in the lungs, and pulmonary hypertension. All results in lower oxygen levels, which is not good.
What about Furin?
Furin is a protein that plays an essential role in the mechanism by which Covid 19 enters lung cells. The process of how Covid 19 interacts with the Furin protein is being investigated as a target site for producing a live vaccine.
So what can we do to restrict the affects of Covid 19 to the upper and conducting pathways and prevent the progression of the virus to other parts of the body that produce more serious symptoms?
